Vector TD is a traditional tracks TD. Desktop TD is one where you create your own tracks, and you can draw your own creeps and stuffz.Rydian said:

I'd say fieldrunners was the most fun, followed by starship patrol and viking invasion.

There was a homebrew one named something along the lines of "Piztog's Tower Defence" which was reasonably decent. And Warcraft Tower Defence as mentioned above is awesome too.
I didn't know desktop tower defence or vector tower defence had DS ports, that's interesting...both homebrew? I'm quite a tower defence fan myself.
