Yes.
Delete the one that downloaded from New Nus Grabber and replace it with the modified one.
FYI, You dont replace update's title.tik's with modified title.tik's. Only add modified title.tik to the full game to be able to install that game. Update's are ready to be installed as is...

Oh I thought it was a full game to be installed, I didn't read it was an update.
If it's an update, then yes you do need to have the one that got downloaded, it's only for FULL games that you replace the tik.

Sorry about the misunderstanding.

Depends.
If you have your game installed on NAND, it should go in NAND, if you have it in your USB, the update should also go to the USB.
In short words, install it to where your game currently is.

it will be USB, but how come i mean i had splatoon installed on USB with wup , and i downloaded the update manually in the WiiU , did that actually download the update onto the USB or NAND?

If you got the download from the eShop I think that's on the NAND.
Check in System Settings to be sure.

Wiiu formated External HDD serves as an extension the internal HDD , so it doesn't matter where you install it ,The game will detect it and use it, i believe.

That's normal... Check your home menu.
It didn't show up on the Menu so I assume it didn't install. Still, for a minute there I thought I had destroyed my Wii U! Twilight Princess and Super Mario 3D world work flawlessly through usb but I wasn't able to install Bayonetta 2 on the internal memory.
 
Ok, so what if I get a game from that i** site, the USB topic one, would I still need NUSGrabber and mod the tickets? Thanks.
There are already people uploading the full nusgrabber wuds with the modded tik file, so you wont need to download the game with nus and deal with its horrible download speed.
