FAT16 is the best option for the R4, as it doesn't support media bigger than 2 GB. By using FAT32, you would lose some space, without any gained advantages.

This is incorrect. FAT32 uses smaller allocation units than FAT; this saves space on the card by wasting less unused space on each file. It's also slower than FAT because more allocation units need to be dealt with for any particular function. Either will work fine on the R4.
