My SD Slot is broken, Can I format a FAT32 flash drive with the same layout to run homebrew?

I already have the Wii U modded. The SD card is no longer functional though and it isn't recognizing USB flash drives as storage (may just need to use an older one though)

I'd like to know if it's even possible, what differences there may be?

No, the slot would have to be repaired. Hard mods seem to be really introducing their way into the Wii U scene so this might make good practice if you are interested.
