1. Can it be used without having a flash card, or is it dependent on any certain type of flash card?

1. You will need a Slot 1 card to actually transfer the GBA games onto the 3 in 1. It's not flash card dependent since there are programs such as the GBAldr that will let you do this.

2. There is no *biggest size*, it allows you to hold 1 game... however I believe it's a 32MB NOR chip.
