Um, I've never played Okami. (I've been planning to get the Wii version but I'm a little strapped for cash recently.)

Will I still be able to enjoy this game?

You'll enjoy it, but some things may go over your head. If anything, it'll make you want to play Okami even more, so I'd go for it.
 
I love this game, but at times there was slow down. At other times, there was even worse slow down. It does not harm the game play, but it is quite annoying at times. Ahem, I am getting off topic.

Okamiden is a most wonderful game and I'll have to skip over to a friend's house and play the original as soon as I can. I may even borrow her Wii so that I can enjoy the game in my own home.

I have never played Okami, but this game is quite enjoyable. It gives you the basic story of Okami in the beginning cutscene (or so I have been lead to believe by a friend of mine), so even if you did not play the first game you will still know what is going on. The story of Okamiden is easy to understand, as well.

I think it's the perfect handheld game to get people into the Okami series.
