DS2x86, the DOS EMU, has been updated for the SCDS2. Please see the change log for more information.

Change Log said:
This version does not have any major new features, mainly just some minor fixes and enhancements compared to 0.01.
• Screen "Scale" mode works also in Mode-X graphics mode.
• The upper and lower screens are swapped when using touchpad mouse.
• Keyboard layout shows upper case characters when in Shift mode.
• Some missing opcodes have been added, based on the log files I have received.
• Screen refresh rate can be changed between 15, 30 and 60fps.
• Lower screen refresh should be more robust than in 0.01 version.
• Lots of protected mode work for DOS4GW, but this work is not yet finished so games using DOS4GW will not work in this version yet.

I don't think so. It uses VGA graphics mode which is not yet supported in DS2x86 and it might run quite slow because the emulator runs at about the same speed as a 40MHz 386 machine, but the game needs a 486/25MHz computer to run smoothly, according to the game description.

Take note that DOS emulation is always a laborious process (see DOSBox) and there are a huge amount of variables to deal with because of the wide variety of programming that goes into DOS games.
There was no real standard in PC programming and so it is a lot more challenging to make an emulator on a DS.
Even so, this emulator is still in its infancy (even though he has made the DSx86 for the standard DS cart.)
There are a lot of differences between programming for the normal DS and programming for the SCDSTWO.

Games work differently in many ways in DOS.
