Open command prompt and type:
format X: /fs:fat32 /size:32k
Not that you need to replace "X" with the actual drive letter of the hard drive you want to format.
Be careful not format the wrong drive!
Either way, yes, you will lose all your data when formatting.

However it appears it's already formatted as fat32, so I don't know if it will make any difference. If there is only one partition on the drive then I doubt it.
