Which is the better way of writing files to the memory stick, usb cable or a MS reader? Or theres no difference?

No difference at all, other than the speed that the data is written/transfered to PSP

Depends on how fast your MS Reader reads. I've never used an MS Reader but the USB Cable connection works pretty fast (plus, all you have to press is O and you're back into your XMB with the newly added files)

I have never bothered with a reader as well

Yup, and a cool, new feature of the Slim is being able to recharge it while it's in USB mode connected through a cable... wewt.
