When Amazon says "No, we won't let you use the Marketplace like everybody else who has an Android device because we like being d*cks", you say "Well, amma gonna pirate all your content and you won't earn jack from me". That's how the world works now.

You can be pretty sure that Amazon will lock down their version of Android to make it purposefully very hard to root.

It can't install APKs, so that approach is gone. Chances are it won't have ADB support (if you can't run custom APKs, who's going to develop on the device?), so that approach is gone. The bootloader is bound to be locked, preventing flashing custom ROMs or update.zips, so that approach is also gone.
