No, they're asking if they could put the DLC for Smash Bros. on the cart somehow, so when it's used in their brother's 2DS it has the DLC. The answer is no, the only part of the cartridge that's writable is the save memory, which would be too small to hold that data.
You can use the dsiwarehax method which will require you to system transfer from your console to his.
I don't think you understand what the OP is trying to do. He is trying to copy the DLC to the actual retail cartridge of Smash which is impossible, CFW or not. ROMs (Read Only Memory) cannot be written to.

Cartridges are read-only. Do you know how to read? You can't write to 3DS carts.
He could either get a compatible ds flash cart with ntr(boot?hax?) and install b9s on his brother's non-hacked 2ds or use the dsiwarehax transfer method.
