I want to update to 4.2.1 but I have a lot of s**t on my ipad. If I will update via Itunes, will I loose everything?
What to do?

If you sync everything before hand and make sure you do a backup everything should be fine

Unless something goes wrong (in which case you will have to restore), updating doesn't remove anything.
