None of those tricks to remove stripped screws (like superglue a screwdriver to the screw, rubber band and such) ever worked for me. In the worst case you will have to carefully drill off the screw heads and remove the back cover. After that pliers can be used in order to turn the remaining parts out (new screws obviously needed).
After that it is possible to clean the inner parts of the button – or – solder in a new button to the cable – or – replace the whole cable with SD reader and shoulder buttons.

In most cases it is corrosion inside the micro switch.Unless I'm mistaken (never had a DSi in my hands) these Micro-Switches tends to break with heavy use/hard presses.
Chances are, your R Button Micro-Switch is loose/broken/desoldered.
