I just copy a game file to sd card and insert back to NS, the console was unabled to boot up then I plugged it back to PC reader and found that the sdcard was corrupted. . .I have to reformat it.

I had power off to insert / remove it from PC. How do I prevent this happen again. . .my card is not fat32.512gb

All the save is lost? I use the sd card as emunand.

Format card to fat32. Exfat is prone to corruption with homebrew. There really is no reason to use exfat.
1)install games via usb
2)ftp small files (less than a gb)
3) :sx os only: play xci games off external hard drive or even install games off hdd
 
Problem on fat32 is need to split 4gb files.

1) you mean connect NS with USB and transfer on win, like read it as card reader?

No no no. The split is only needed if you were to transfer it to sd card to install. What I'm saying is leave the nsp/xci/nsz on ur computer, plug in you NS the computer and install directly to you NS from pc. You dont need to transfer to sd to install, dont need to split files bigger than 4gb to install. Heck, drag and drop the games (nsp/xci/nsz) to root of a external hdd, or thumb drive (flash drive, whateveryou wanna call it), plug the external hdd (or flash drive) to the dock, put NS in dock and you can install the games from external hdd.
