Did you reinstall the IOS needed for WiiFlow, USB Loader GX and NeoGamma after your updated your Wii? The update will probably have overwritten some of them with stubs.

Which version did you install ? (this question for both cIOS and loader)
To be sure you have everything correctly installed, you can use modMii to download all the required files and generate a personal tutorial.

You need to install this (look at all the details I provide) :

cIOS d2x version 10 based on IOS 56 installed in slot 249
cIOS d2x version 10 based on IOS 57 installed in slot 250

if you want USBLoaderGX, the latest release is revision 1215. Use the manual installation, don't use the channel (which is revision 1180)
    Wiiflow, I don't know latest revision/version
    Neogamma is old

Well where is your USB loader installed? On the SD card or on your USB drive?

Not really, but you can install it to your USB HDD you need anyway, if it is formated with FAT or NTFS. It will work without the SD card then.

yes, you can install homebrew on your Wii instead of the SD or USB.
For example, USBLoaderGX is available in two different mode : Full channel (installed on Wii), or Forwarder channel/boot.dol for SD or USB.

Though, you will still need a device to store the settings, game covers, etc.
So you should use the Forwarder channel and place the boot.dol on your USB if you don't have SD card. it will be easier to update when new versions are released (just replace the boot.dol on usb).

Oh ehm did not think of this, sorry for the slight mis-information.
But as you still need a device for storage for covers, config, etc like Cyan said, I think that this is pretty useless.

Some games doesn't work with USBloaders. Tintin is one of them.

Tintin works only when booted from System menu.
You can use Sneek (please, use the search feature of the forum and read what you find before asking question about sneek) to play that game in ISO/FST backup format, or using a modchip/patched IOS is you want to play from Disc backup.
Easier method for Tintin is to use retail game from system menu.
