Diddy Kong Racing has one of the best single-card multiplayer offerings. It's been a while, but if memory serves, you get access to all the characters, tracks and vehicles (though there are only three of those) you've unlocked in the single-player game.

Worms OW2 also has a great single-card option.

My personal favorites are.

Mario Party DS (Alot of fun and alot of stuff to play with 1 cart)

Clubhouse Games/42 all time classics (You can play I believe 39 of the games on ds download play single cart. You can play them online also)
