A: Yes. However for DS game, the reading speed is not fast enough, so, DS games don’t support Exfat. But if you play 3DS only, you can use Exfat.

This mean that DSTWO PLUS can't save on Card2 games like Animal Crossing, Pokemon...?

This is broken english, but to me it sounds like it's telling you to use a clean rom and not rip your own rom from your legit cartridges.
 
Well from what I can analyse from the engrish language, I guess that it means to use a Clean / Untrimmed ROM.
i.e. for Pokemon games, you should use the full 2 GB ROMs and not a trimmed one which is like 1.4 or so GB I think.
