Almost yeah, i used a custom "Toony colors pro" shader for skin and cloth but you can easily reproduce it with amplify (i will later ;)). Imo, ASE is the most powerfull tool for shaders in Unity.

Like Manurocker said, with photoshop you can load .dds and save them as .png, be sure to allow Alpha if the texture need it (usually photoshop ask for it if you import an alpha texture).
