How can I patch a full size iso or wbfs with patch meant to be used with Riivolution?
Is there any tool that can help me achieve this?

Does anybody know about the tools to inject ROMs into Wii Virtual Console wads? Do
we have any compatibility list for this?

The wiki section of GBATemp should have a VC Injection compatibility page. Also, there's a tutorial in Wii - Tutorials for that.
And no, Riivolution does not work with USB/DVD backups. You can try using something like Dolphinlution if you want to make ISOs for Riivolution packs.
