Do I need some sort of special cable (like an eSATA to USB) plugged from the HDD to the Wii for it to run games/movies optimally? Is that even possible? Would a straight USB to USB connection work just fine without any noticable slowdowns? Am I somehow over thinking this a little?

Okay, given that, would an eSATA to USB work even better?

No, because you're not removing any steps - you're still connecting the drive to some sort of enclosure or adapter via SATA and then to the Wii via USB. eSATA/USB adapters aren't any faster than HDD enclosures.

You can use either, but there's no performance improvement.
