No, unless you have a modchip, and use WUM to patch the iso with the latest update. if you using backup launchers theres no chance of doing it without internet connection m8.

I think you can update to 3.2 using these special update disks: http://hackmii.com/2008/05/semi-brick-fix-discs/
If the links are dead just scroll down for mirrors

You can probably burn the iso and launch with backup loader or extract the system update wad from the iso using wii scrubber and install that using a wad manager. I havent tested it myself but I don't see why it wouldn't work
