only with homebrew compatible with HID-USB controllers.
nintendont for sure.
maybe some loaders (wiiflow?) but only while inside that homebrew, not when playing wii games.
maybe retroarch as it's still being active? I don't know.
all old homebrew before that controller was released will not be compatible.
new ones, only if support was specifically added, it's not a real native gamecube controller port.

I think a few of the early systems could benefit from that, at least up to the 8 bit generation, by the 16 bit generation I think TVs had gotten a lot better in almost every way

Just give it a try, but honestly if you have a 3DS you can play DS games without a card just off the internal SD card
