I am thinking of buying Animal Crossing. Will it be playable through HomePass? I mean visiting other Miis etc or not?

There are reports that you get sp from others with AC. So yes you can visit others home via homepass.

Has anyone confirmed it? Do I have to run ACNL while my 3DS is in sleep (streetpass) mode?

I can. It doesn't have to be in sleep mode.
